แมรี-หลุยส์ ปาร์กเกอร์ เกิดในเมือง Fort Jackson, เซาท์แคลโรไรนา สหรัฐอเมริกา เป็นลูกคนเล็กของพี่น้องสี่คน, แม่ของเธอ แคโรไลน์ หลุยส์ (สกุลเดิม Morell) พ่อ จอห์น มอร์แกน ปาร์กเกอร์ เป็นผู้พิพากษาที่เคยทำงานในกองทัพบกสหรัฐ [1][2][3] บรรพรุษของเธอ เป็นคนสวีเดน (จากปู่ของแม่) อังกฤษ สก๊อต สก๊อต-ไอริช เยอรมัน และดัทช์[4] เนื่องจากการทำงานพ่อของเธอ ปาร์กเกอร์ก็ได้ใช้เวลาในวัยเด็กใน เทนเนสซี แล้ะเท็กซัส และในประเทศไทย เยอรมนี และฝรั่งเศส[5][6] เธอเล่าชีวิตวัยเด็กของเธอว่า "ไม่มีความสุขอย่างยิ่ง"  "พ่อแม่ของฉันทำทุกอย่างที่สามารถทำได้ ฉันมีทุกอย่าง หนังสือ เสื้อผ้า บ้าน และเตียงที่อบอุ่น แต่ไม่เคยมีความสุข"[1] เธอเรียนจบจากโรงเรียนมัยมตอนปลาย Marcos de Niza ในเมือง Tempe, รัฐแอริโซนา ปาร์กเกอร์สำเร็จการศึกษา สาขาการละคร จากมหาวิทยาลัยนอร์ทแคโรไลนา ในปี 1986

ในช่วงปี 1997 จนถึงเดือนพฤศจิกายน ปี 2003 ปาร์กเกอร์เคยมีความสัมพันธ์กับนักแสดง Billy Crudup แต่เขาก็ทิ้งเธอไปหาแคลร์ เดนส์ ขณะที่เธอตั้งครรภ์ได้เจ็ดเดือนกับลูกของเขา, William Atticus Parker (เกิดในปี 2004)[7] นักแสดง ซูเซิน ซาแรนดอน ให้เกียรติรับเป็นแม่อุปถัมภ์ให้กับลูกชายของเธอ

ในธันวาคม ปี 2006 ปาร์กเกอร์เริ่มเดทกับนักแสดง Jeffrey Dean Morgan ซึงพบกันที่กองถ่ายภาพยนตร์ Weeds[8] ในเดือนกุมภาพันธ์ ปี 2008 เธอและมอร์แกนประกาศการหมั้น[9] แต่ก็เลิกจากกันในเดือนเมษายน[10]

ในกันยายน ปี 2007 ปาร์กเกอร์รับเลี้ยงเด็กบุญธรรม Caroline Aberash Parker จากเอธิโอเปีย[11][12]

ในปี 2013 ปาร์กเกอร์ได้รับเกียรติจาก Hope North ซึ่งเป็นองค์การให้การศึกษาและการรักษาเด็กๆ จากสงครามกลางเมืองในอูกันดา ปาร์เกอร์เริ่มมีส่วนเกี่ยวข้องกับองค์กรดังกล่าวหลังจากได้พบปะกับอดีตเหยื่อคนหนึ่งจากสงครามกลางเมืองอูกันดา[13]
