สตรีหมายเลขหนึ่ง (อังกฤษ: first lady) เป็นตำแหน่งอย่างไม่เป็นทางการที่มักใช้กับภรรยาของประธานาธิบดีหรือประมุขแห่งรัฐที่ไม่ใช่พระมหากษัตริย์ (ในบางโอกาสใช้แก่ลูกสาวหรือญาติฝ่ายหญิงคนอื่น ๆ)[1][2][3] คำนี้ยังสามารถใช้ระบุสตรีที่มีอาชีพหรือศิลปะอยู่ในอันดับต้น ๆ ด้วย[4]
ตำแหน่งนี้สามารถใช้กับภรรยาของหัวหน้ารัฐบาลที่ไม่ได้เป็นประมุขแห่งรัฐ[5][6][7][8] และสามารถสื่อถึงภรรยาของหัวหน้าเขตบริหารในประเทศได้เช่นกัน[9]
