แซม คุก (อังกฤษ: Sam Cooke) หรือแซมูเอล คุก (อังกฤษ: Samuel Cook)^[4] (22 มกราคม ค.ศ. 1931-11 ธันวาคม ค.ศ. 1964) เป็นนักร้องแนวกอสเปล อาร์แอนด์บี ดนตรีโซล และป็อป เป็นนักแต่งเพลง ผู้บริหาร ชาวอเมริกัน ถือเป็นผู้บุกเบิกรุ่นแรก ๆ ของวงการเพลงโซล^[5]^[6]^[7]

คุกมีเพลงฮิตติดท็อป 40 ในอเมริกา 29 เพลง ในระหว่างปี 1957-1965 มีเพลงดังสำคัญ ๆ อย่าง "You Send Me", "A Change Is Gonna Come", "Chain Gang", "Wonderful World" และ "Bring It on Home to Me" คุกยังเป็นนักร้องและนักแต่งเพลงผิวดำคนแรก ๆ ที่ก้าวสู่วงการธุรกิจ เขาก่อตั้งค่ายเพลงและบริษัทสิ่งพิมพ์ เพื่อขยับขยายงานทางด้านร้องเพลงและแต่งเพลงของเขา ทางด้านงานสังคมเขายังเป็นส่วนหนึ่งของ American Civil Rights Movement^[8]
